A new simple and highly selective and sensitive catalytic differential pulse voltammetry procedure for the determination of thiourea at nanomolar level is reported. Thiourea has a catalytic effect on the oxidation of Janus green by iodate in the hydrochloric acid medium. The potential was scanned in the negative direction and the differential pulse voltammograms were recorded. The variations of the peak current with hydrochloric acid concentration, oxidant, Janus green, pulse amplitude, pulse time and scan rate were optimized. Under the optimized conditions, the relationship between the peak current and concentration of thiourea was obtained. It is shown that the calibration curve is linear in the range of 0.01–6.00 µg/mL. The detection limit of the method was 0.005 µg/mL. The relative standard deviation for 6 replicate determinations of 0.01, 0.50 and 2.00 µg/mL is equal to 2.25%, 1.52% and 1.03%, respectively. The method was applied to the determination of thiourea in fruit juices with satisfactory results.  相似文献

提出了应用气相色谱-串联质谱法测定蔬菜(西红柿、黄瓜等)和水果(橙子、苹果等)中潜在的污染物三氯生(TCS)和甲基三氯生(MTCS)的含量的方法,以期有助于对此污染物在蔬菜、水果等食品中的污染问题的研究,并应用于农产品质量的安全监测之中。将样品分别去皮后切块,匀浆后于-22℃保存。称取样品5.00g,先后2次用乙酸乙酯(每次10mL)超声提取10min后离心10min,合并2次的提取液。将提取液吹氮蒸发至近干。加入衍生试剂体积比为99∶1的N,O-双(三甲基硅烷基)三氟乙酰胺-三甲基氯硅烷混合溶液100μL,在60℃下进行衍生化反应40min。反应结束后,将溶液吹氮至近干,用乙酸乙酯1.0mL溶解残渣。所得溶液供气相色谱-串联质谱法分析。用DB-17MS毛细管色谱柱(30m×0.25mm,0.25μm)在150~290℃区间按程序升温条件进行分离,在电子轰击离子源及多反应监测模式下进行质谱测定。经试验确定,用两种扫描程序分别扫描,TCS衍生物的母离子和子离子依次为m/z 362,347和m/z 360,345,MTCS的母离子和子离子则均为m/z 304,254。测得TCS衍生物和MTCS的标准曲线的线性范围均为0.4~20.0μg·L^-1,其检出限(3S/N)为4~6ng·kg^-1。按标准加入法进行回收试验,测得TCS衍生物和MTCS在4种蔬菜、水果中的回收率为91.5%~107%和94.7%~111%。  相似文献

In general, lignocellulosic biomass contains three major components, namely lignin, hemicellulose and cellulose which are the polymers of C5 and C6 sugars. Thus, there is potential to utilize of this biomass for bioethanol production. The hydrolysis of cellulose into glucose was difficult due to the more fibrous nature and thus inhibit enzyme penetration into the cellulose. In order to solve this problem, hydrothermal pretreatment can be used for breaking the bonds within the lignin structure and increase the accessibility of enzyme into the cellulose. In this study, the effect of chemical addition, sodium hydroxide (NaOH) and calcium oxide (CaO) in hydrothermal pretreatment at 180 °C and 30 minutes reaction time of palm oil empty fruit bunches (EFB) on the enzymatic hydrolysis efficiencies was investigated. The enzymatic hydrolysis of hydrothermally pretreated EFB give the highest concentration of glucose at 0.67 g/L while the hydrothermally pretreated of EFB in the presence of NaOH gives the lowest glucose concentration 0.45 g/L.  相似文献

Gellan was used to suspend pulp particles in orange juice. Three groups of samples were prepared with 0%, 20%, and 40% orange juice concentrate and supplemented with gellan at different concentrations. A concentration-dependent increase in the size of gellan aggregates and gellan-protein assemblies was observed. Incorporation of gellan into the beverage with 0% juice concentrate changed the rheological behavior of sample to non-Newtonian shear-thinning fluid and increased its surface tension. When juice concentrate proportion was increased from 0% to 20%, the beverage viscosity increased. The highest gellan concentration resulted in a higher yield stress (σ0) value and inhibited the pulp sedimentation completely.   相似文献

A stir bar sorptive extraction method coupled with deep eutectic solvent based solidification of floating organic droplets–dispersive liquid–liquid microextraction has been used for the simultaneous derivatization and extraction of some acidic pesticides in tomato samples. In this method, initially the analytes are adsorbed on a coated stir bar from tomato juice filled in a narrow tube. After extraction, the stir bar is removed and a water–miscible deep eutectic solvent is used to elute the analytes. Afterward, a derivatization agent and a water–immiscible deep eutectic solvent (as an extraction solvent) with melting point near to room temperature are added to the obtained eluant at µL–levels and the obtained mixture is rapidly injected into deionized water. Under the optimum conditions, the introduced method indicated high enhancement (1543–3353) and enrichment (2530–2999) factors, low limits of detection (7–14 ng/L) and quantification (23–47 ng/L), good linearity (r2 ≥ 0.9982), and satisfactory repeatabilities (relative standard deviation ≤12% for intra– and inter–day precisions at a concentration of 100 ng/L of each analyte). Finally, the proposed method was applied in analysis of the analytes in tomato samples.  相似文献

A simple, selective, sensitive and inexpensive method of hollow fiber-based liquid–liquid–liquid microextraction (HF-LLLME) combined with high performance liquid chromatography (HPLC)-ultraviolet (UV) detection was developed for the determination of four acidic phytohormones (salicylic acid (SA), indole-3-acetic acid (IAA), (±) abscisic acid (ABA) and (±) jasmonic acid (JA)) in natural coconut juice. To the best of our knowledge, this is the first report on the use of liquid phase microextraction (LPME) as a sample pretreatment technique for the simultaneous analysis of several phytohormones. Using phenetole to fill the pores of hollow fiber as the organic phase, 0.1 mol L−1 NaOH solution in the lumen of hollow fiber as the acceptor phase and 1 mol L−1 HCl as the donor phase, a simultaneous preconcentration of four target phytohormones was realized. The acceptor phase was finally withdrawn into the microsyringe and directly injected into HPLC for the separation and quantification of the target phytohormones. The factors affecting the extraction efficiency of four phytohormones by HF-LLLME were optimized with orthogonal design experiment, and the data was analyzed by Statistical Product and Service Solutions (SPSS) software. Under the optimized conditions, the enrichment factors for SA, IAA, ABA and JA were 243, 215, 52 and 48, with the detection limits (S/N = 3) of 4.6, 1.3, 0.9 ng mL−1 and 8.8 μg mL−1, respectively. The relative standard deviations (RSDs, n = 7) were 7.9, 4.9, 6.8% at 50 ng mL−1 level for SA, IAA, ABA and 8.4% at 500 μg mL−1 for JA, respectively. To evaluate the accuracy of the method, the developed method was applied for the simultaneous analysis of several phytohormones in five natural coconut juice samples, and the recoveries for the spiked samples were in the range of 88.3–119.1%.  相似文献

Organic acids, sugars, phenolic compositions and antioxidant capacities of orange juice and orange wine obtained from the cv. Kozan of Turkey were determined. High-performance liquid chromatographic methods were used to identify and quantify of these compounds. Three organic acids (citric, malic and ascorbic acids) and three sugars (sucrose, glucose and fructose) were determined. The major organic acid was found as citric acid. With regard to sugars, sucrose was present in the largest amounts for orange juice and wine. A total of 13 phenolic compounds were identified and quantified in orange juice and wine, including hydroxybenzoic acids (2), hydroxycinnamic acids (5), and flavanones (6). Hesperidin, narirutin and ferulic acid were the most abundant phenolic compounds in orange juice and wine. Antioxidant activities of orange juice and wine were measured using the DPPH (2,2-diphenyl-1-picrylhydrazyl) assay, and the antioxidant capacity of orange juice was found to be higher than that of orange wine.  相似文献

Typical flavour of Muscat d’Alexandrie wines is mainly due to volatile compounds coming from grapes of this variety. Therefore, the choice of grapes is crucial to obtain musts with a great aromatic potential, which will contribute to the final aroma of wines derived from those musts. In this study, three sampling techniques, liquid-liquid extraction (LLE), solid phase extraction (SPE) and simultaneous distillation-extraction (SDE), were compared for the analysis of volatile compounds in Muscat grape juice. Results showed that although the three techniques can be recommended for the quantitative analysis of volatile compounds from musts, LLE and SPE are better sample preparation techniques than SDE, mainly for determination of polar compounds such as acids or alcohols.  相似文献
